WS Biotin™ is water-soluble D-Biotin (50% w/w, 2.2g/100ml) for skin care; at 0.1% it cut UVA-induced melanin by 18.4% in vitro, with no crystallization.

WS Biotin™ for Skin Care: At Last, an Easy Way to Design Highly Concentrated, Effective D-Biotin Formulations

D-Biotin and Its Formulation Challenges

Biotin is a common supplement, also known as vitamin B7. It plays a critical role in a handful of metabolic processes in our body. More specifically, Biotin helps improve skin's pigmentation, hydration, smoothness, and appearance. Biotin occurs as a practically white, crystalline powder with very little solubility in water, alcohol and other common organic solvents. Therefore, there is a real need to increase its water solubility to overcome this formulation challenge, allowing the design and development of effective skin products.

Tagra's Solution

To improve D-Biotin's effectiveness in skin care formulations, Tagra has developed a groundbreaking, innovative solution: WS Biotin™ is a water-soluble, dry powder form of D-Biotin (50% w/w), offering a higher solubility level of 2.2g/100ml compared to 0.02g/100ml for free D-Biotin. Tagra's unique technology leads to a significant increase in aqueous solubility without high-energy investment, showing WS Biotin™ to be highly useful.

How Do We Know It Works?

Melanin is primarily responsible for the pigmentation of human skin, eyes and hair. Among other stimuli, ultraviolet radiation triggers the biosynthesis of melanin and the subsequent melanosome transfer to epidermal keratinocytes.

We assessed the inhibitory capabilities of WS Biotin™ on melanin biosynthesis through a 72-hour in vitro treatment on Normal Human Epidermal Melanocytes (NHEM) that were subjected to ultraviolet A (UVA) irradiation, followed by a 48-hour period of intracellular melanin accumulation.

Results obtained from cells co-exposed to UVA and WS Biotin™ for 72 hours strongly suggested that this product at 0.1% displays whitening activity, demonstrated by a reduction of UVA-induced melanin levels by 18.4% compared to the untreated Control + UVA.

Effect of WS Biotin™ on the UVA-induced accumulation of melanin: intracellular melanin levels after 3 days of simultaneous exposure to UVA (30 J/cm²) and treatment with WS Biotin™, followed by 2 days of melanin accumulation in the presence of WS Biotin™ in Normal Human Epidermal Melanocytes. Groups: Control, Control + UVA, 0.1% WS Biotin™.

How to Formulate with WS Biotin™

WS Biotin™ is a potent and practical solution for skincare (serum, creams, gel) products. Formulating with WS Biotin™ results in a pH of 6-7 and does not require using any organic solvents for solubilizing D-Biotin. It prevents any risk of crystallization in the formula, and it is added to the formulation at room temperature. No mixing process is required as WS Biotin™ is solubilized spontaneously.

Water Solubility of WS Biotin™ in Comparison to Free Biotin

  • Left: WS Biotin™ water solution at 4% (equivalent to 2% free D-Biotin) after 30 days incubation at 10°C.

  • Right: 0.1% water solution of free D-Biotin after 24h incubation at 10°C.

Crystals were formed in the free D-Biotin solution while no crystals were formed in the WS Biotin™ water solution.
